I want to use this ritual against an enemy. I want them not only to lose physical beauty and attraction power, but also that their horrible personality is seen by everyone around them.
Is it possible that during my petition I ask for these two things or should I focus on one?
Yes, you can use the one ritual as these are so closely related, I don’t see an issue with muddying the intention. Overall you want them to be seen as an ugly person, in every way.

You can, but I would not. Having used this book a few times, I didn’t start getting results with it until I really spend time and energy on the ritual, and doing them separately would help you get that level of effort in.
The time I found I connected best with it, I actually did the ritual while on a run on a treadmill. This kind of intense training puts the mind in a kind of trance that is perfect for really getting repetitive and focused on one thing - the body is occupied and the mind can get bored, but not if you use that freed up mental energy for magick. I ran for an hour and focused on the Exile working the entire time.
(It sort of worked, but it’s blocked, so the energy leaked out and hit in every other way it could, three or four neighbors that I don’t care about all moved instead 
but it’s powerful - when you put the time in.)
So I would say, don’t skimp on this, give it more than you feel is really necessary and it’s worth it.

I performed the Destroy Defenses ritual for both of them as a unit but following this I suppose I would have to do it again for each person?
